The cultivation of summer vegetables in open-air nutrient film technique (NFT) hydroponics is limited due to the elevated nutrient solution temperature (NST). In this regard, non-electric evaporative-cooling techniques were explored to maintain NST in open-air NFT hydroponics. Four cooling setups were employed by wrapping polyvinyl chloride (PVC) grow pipes with one and two layers of either wet or dry jute fabrics and attaching them with coiled aluminum pipe buried inside a) wet sand-filled brick tunnels (Cooling Setup I), b) two inverted and vertically stacked earthen pots (Cooling Setup II), c) two inverted and vertically stacked earthen pots externally wrapped with wet jute fabric (Wrapped Cooling Setup II), and d) an earthen pitcher wrapped with wet jute fabric (Cooling Setup III). Wrapping grow pipes with two layers of wet jute fabric reduced NST by 5°C as compared to exposed (naked) grow pipes. The double-layer jute fabric-wrapped grow pipes produced 182% more reduction in NST in comparison to single layer-wrapped grow pipes. Additionally, the installation of Wrapped Cooling Setup II and Cooling Setup III outperformed Cooling Setup I and Cooling Setup II through NST reduction of approximately 4°C in comparison to control. Interestingly, Cooling Setup III showed its effectiveness through NST reductions of 193%, 88%, and 23% during 11 a.m.-12 p.m. as compared to Cooling Setup I, Cooling Setup II, and Wrapped Cooling Setup II, respectively. In contrast, Wrapped Cooling Setup II caused NST reductions of 168%, 191%, and 18% during 2-3 p.m. in comparison to Cooling Setup I, Cooling Setup II, and Cooling Setup III, respectively. Thus, the double-layer jute fabric-wrapped grow pipes linked with Wrapped Cooling Setup II can ensure summer vegetable cultivation in open-air NFT hydroponics as indicated by the survival of five out of 12 vegetable plants till harvest by maintaining NST between 26°C and 28°C.

The impact of baffles on a convective heat transfer of a non-Newtonian fluid is experimentally studied within a square cavity. The non-Newtonian fluid is pumped into the cavity through the inlet and subsequently departs from the cavity via the outlet. Given the inherent non-linearity of the model, a numerical technique has been selected as the method for obtaining the outcomes. Primarily, the governing equations within the two-dimensional domain have been discretized using the finite element method. For approximating velocity and pressure, we have employed the reliable P 2 - P 1 finite element pair, while for temperature, we have opted for the quadratic basis. To enhance convergence speed and accuracy, we employ the powerful multigrid approach. This study investigates how key parameters like Richardson number (Ri), Reynolds number (Re), and baffle gap b g influence heat transfer within a cavity comprising a non-Newtonian fluid. The baffle gap ( b g ) has been systematically altered within the range of 0.2-0.6, and for this research, three distinct power law indices have been selected namely: 0.5, 1.0, and 1.5. The primary outcomes of the investigation are illustrated through velocity profiles, streamlines, and isotherm visualizations. Furthermore, the study includes the computation of the Nu avg (average Nusselt number) across a range of parameter values. As the Richardson number (Ri) increases, Nu avg also rises, indicating that an increase in Ri results in augmented average heat transfer. Making the space between the baffles wider makes heat flow more intense. This, in turn, heats up more fluid within the cavity.

In terms of delivery systems for active compounds, orally disintegrating films are a great option. The initial stage in creating an oral disintegrating film is selecting a film-forming polymer. The basic polymers combination Microcrystalline Cellulose (MCC), which is co-processed with Carboxymethylcellulose Sodium (CMC) and hydroxypropylmethyl cellulose were used to create an oral disintegrating film that contains cholecalciferol (Vitamin D3), a fat-soluble vitamin that aids in the body's absorption of calcium and phosphorus. The goal of the current inquiry was to develop orally disintegrating films of vitamin D3 to improve patient comfort and compliance for pediatric or elderly patients due to its simplicity of administration. Films containing drugs and made of the appropriate plasticizer and chosen polymers demonstrated outstanding film forming and folding endurance. The dissolution test showed that Vitamin D3 has a rapid disintegration property, with the majority of it dissolving in the medium (pH 6.8) in less than two minutes after being inserted. To verify that the films were successfully formed, a variety of procedures including HPLC, FT-IR and microscopic studies were employed. When kept at 40oC with humidity of 75%, the film showed good stability for at least three months.

Amongst hip fracture admissions, mortality is higher in men than in women. However, sex differences in other care-quality measures have not been well-documented. We aimed to examine sex differences in mortality as well as a wide range of underlying health indicators and clinical outcomes in adults ≥ 60 year of age admitted with hip fractures from their own homes to a single NHS hospital between April-2009 and June-2019. Sex differences in delirium, length of stay (LOS) and mortality in hospital, readmission, and discharge destination, were examined by logistic regression. There were 787 women and 318 men of similar mean age (± SD): 83.1 year (± 8.6) and 82.5 year (± 9.0), respectively (P = 0.269). There were no sex differences in history of dementia or diabetes, anticholinergic burden, pre-fracture physical function, American Society of Anesthesiologists grades, or surgical and medical management. Stroke and ischaemic heart disease, polypharmacy, and alcohol consumption were more common in men. After adjustment for these differences and age, men had greater risk of delirium (with or without cognitive impairment) within one day of surgery: OR = 1.75 (95%CI 1.14-2.68), LOS ≥ 3 weeks in hospital: OR = 1.52 (1.07-2.16), mortality in hospital: OR = 2.04 (1.14-3.64), and readmission once or more after 30 days of a discharge: OR = 1.53 (1.03-2.31). Men had a lower risk of a new discharge to residential/nursing care: OR = 0.46 (0.23-0.93). The present study revealed that, in addition to a greater risk of mortality than women, men also had many other adverse health outcomes. These findings, which have not been well-documented, serve to stimulate future targeted preventive strategies and research.

Soil urease inhibition slows down the urea hydrolysis and prolongs nitrogen (N) stay in soil, resulting in an increased N uptake by plants. Apart from several chemical urease inhibitors, the urease inhibition potential of plant extracts is rarely reported. In our previous study, the soil urease inhibition by Vachellia nilotica leaf extract was reported; however, its role in relation to growth and yield of wheat (Triticum aestivum) under pot and field conditions remains unknown. The acetonic extracts of 10, 20, and 50 g Vachellia nilotica leaves were given code names viz. Vn.Fl-10, Vn.Fl-20 and Vn.Fl-50, respectively, and coated on 100 g of urea individually. The enhancements of growth (total number of tillers, number of productive tillers, number of spikelets per spike, number of grains per spike, and 1000-grains weight) and yield (biological yield, straw yield, and grain yield) parameters of wheat by Vn.Fl-20 and Vn.Fl-50 coated urea treatments were compared with uncoated urea in a pot experiment. The experiment indicated that the Vachellia nilotica extract coatings were effective at improving N persistence in soil, as reflected by increased grain and straw N concentrations as well as uptakes. The reproduction of the aforementioned results, at the half and full recommended dose of urea under field conditions, reconfirmed the effectiveness of Vachellia nillotica coatings. Moreover, the Vn.Fl-20 and Vn.Fl-50 coated urea, at the half as well as full recommended dose under field conditions, proved equally effective in terms of higher biological, straw, and grain yield, and grain N uptake. The increments in the total number of tillers, number of productive tillers, 1000-grain weight, biological yield, straw yield, grain yield, grain N concentration, grain N-, and straw N uptake along with nitrogen use efficiency (NUE) components, i.e. nitrogen partial factor productivity (NPFP), nitrogen agronomic efficiency (NAE), partial nitrogen balance (PNB), and nitrogen recovery efficiency (NRE) of wheat highlighted the superiority of Vn.Fl-20 coating over the hydroquinone (Hq) coating on urea at the full recommended dose under field conditions. Given the findings of this study, Vachellia nilotica leaf extract coating (Vn.Fl-20) can be used as a natural urease inhibitor to reduce urea hydrolysis and enhance wheat productivity.

The reliability of the usage of a splitter plate (passive control device) downstream of the obstacle, in suppressing the fluid forces on a circular obstacle of diameter [Formula: see text] is studied in this paper. The first parameter of the current study is the attachment of a splitter plate of various lengths [Formula: see text] with the obstacle, whereas the gap separation [Formula: see text] between the splitter plate and the obstacle, is used as a second parameter. The control elements of the first and second parameters are varied from [Formula: see text] to [Formula: see text]. For the attached splitter plates of lengths [Formula: see text] and [Formula: see text], the oscillatory behavior of transient flow at [Formula: see text] is successfully controlled. For the gap separation, [Formula: see text] and [Formula: see text] similar results are obtained. However, it is observed that a splitter plate of too short length and a plate located at the inappropriate gap from the obstacle, are worthless. A computational strategy based on the finite element method is utilized due to the complicated representative equations. For a clear physical depiction of the problem, velocity and pressure plots have been provided. Drag and lift coefficients the hydrodynamic benchmark values are also evaluated in a graphical representation surrounding the obstacle's peripheral surface as well as the splitter plate. In a conclusion, a splitter plate can function to control fluid forces whether it is attached or detached, based on plate length and gap separation between obstacle and plate, respectively.

Nitrogen (N) deficiency is a primary limiting factor for crop production worldwide. Previously, we reported root system architectural modifications of hydroponically cultured foxtail millet [Setaria italica (L.) Beauv.] to facilitate N translocation under N limitation. Here, we investigated foxtail millet for its shoot adaptation to low N in terms of internal N regulation under hydroponic culture. The results of this study revealed that the shoot N and nitrate (NO3-) concentrations significantly declined as compared to control (CK); however, the shoot over-accumulated ammonium (NH4+) under low N (LN). N shortage resulted in down-regulation of expressions of SiPetA, SiccsA, SipsbA, SirpoB, SipsaA, SiatpA, Sirps16, and SiPEPC which, undermined chloroplast functioning and CO2 assimilation for the provision of carbon skeleton. Carbon deficiency and lower activities of GS decelerated ammonia assimilation and led to over-accumulation of NH4+ in the LN-shoot, as indicated by lower concentrations of total amino acids. Thus, enhanced GOGAT activity was to assimilate NH4+ while, those of catalase (CAT), superoxide dismutase (SOD) and peroxidase (POD) were to scavenge reactive oxygen species (ROS) of NH4+ toxicity framework. The weakened chloroplast factory eventually minimized photosynthesis and reduced dry mass of the LN shoot. Such regulation of N by the shoot, perhaps, resurrected physiological functions which maintained internal mineral status under nitrogen limitation in foxtail millet.

In the current work, an investigation has been carried out for the Bingham fluid flow in a channel-driven cavity with a square obstacle installed near the inlet. A square cavity is placed in a channel to accomplish the desired results. The flow has been induced using a fully developed parabolic velocity at the inlet and Neumann condition at the outlet, with zero no-slip conditions given to the other boundaries. Three computational grids, C1, C2, and C3, are created by altering the position of an obstacle of square shape in the channel. Fundamental conservation and rheological law for viscoplastic Bingham fluids are enforced in mathematical modeling. Due to the complexity of the representative equations, an effective computing strategy based on the finite element approach is used. At an extra-fine level, a hybrid computational grid is created; a very refined level is used to obtain results with higher accuracy. The solution has been approximated using P2 - P1 elements based on the shape functions of the second and first-order polynomial polynomials. The parametric variables are ornamented against graphical trends. In addition, velocity, pressure plots, and line graphs have been provided for a better physical understanding of the situation Furthermore, the hydrodynamic benchmark quantities such as pressure drop, drag, and lift coefficients are assessed in a tabular manner around the external surface of the obstacle. The research predicts the effects of Bingham number (Bn) on the drag and lift coefficients on all three grids C1, C2, and C3, showing that the drag has lower values on the obstacle in the C2 grid compared with C1 and C3 for all values of Bn. Plug zone dominates in the channel downstream of the obstacle with augmentation in Bn, limiting the shear zone in the vicinity of the obstacle.

Fluoride (F) is an emerging pollutant that originates from multiple sources and adversely affects plant growth and nutrient bioavailability in soil. This greenhouse study investigated the effects of soil F (0, 10, 20, 50, 100, 200 mg kg-1) on morpho-physiological growth characteristics of wheat, soil F contents, and bioavailability and uptake of F, phosphorus (P), sulphur (S), potassium (K), calcium (Ca), magnesium (Mg), aluminium (Al), iron (Fe), manganese (Mn), silicon (Si) and zinc (Zn) by wheat. Higher F significantly reduced plant height and number of leaves particularly at early growth stages and increased visible leaf injury index. Powdery mildew infestation coincided with leafy injury and was higher in elevated soil F treatments. Fluoride treatments (>50 mg kg-1) significantly increased water (H2O)- and calcium chloride (CaCl2)-extractable F contents in soil. Water-extractable soil F contents from soil in all concentration were higher than CaCl2-extractable F. This increased F bioavailability resulted in significantly higher F uptake and accumulation in live leaves, dead leaves and grains of wheat which followed order: live leaves > dead leaves > grains. Leaf injury index and number of dead leaves correlated significantly positively with soil H2O- and CaCl2-extractable F contents. Patterns of nutrient (P, K, S) and trace metals (Al, Ca, Mg, Fe, Mn, Si, Zn) varied significantly with F concentrations and between live and dead leaves, and grains except for Zn. Dead leaves generally had higher nutrients and trace metals than live leaves and grains. Fluoride contents in live leaves, dead leaves and grains showed positive correlations with nutrient elements but negative with trace metals. Number of dead leaves correlated negatively with Al, Ca, Fe, Mg, S and Si but positively with P and Zn contents in dead leaves whereas leaf injury index showed positive correlation with Fe, K, P, Si, Zn, S but negative with Al, Ca and Mg contents. These observations provided evidence of higher F uptake and associated impairment in nutrient and trace metal accumulation which caused leaf injury accompanied by powdery mildew infestation in wheat. However, further research in the region is required to confirm the relationship between F pollution, leaf injury and trace metal accumulation in crops under field conditions.

Honey bees (Apis mellifera) are essential for their products-honey, royal jelly, pollen, propolis and beeswax. They are also indispensable because they support ecosystems with their pollination services. However, the production and functions of honey bees are hindered by the arthropod pest Varroa destructor, which attacks bees through its feeding activities. Efforts to control varroa mites have been made through the development of various synthetic pesticide groups, but have had limited success because the mites developed resistance and some of these pesticides are harmful to bees. Branded pesticides are rarely used in Pakistan, as beekeepers utilize acaricides from unknown sources. There is a need to create awareness of available naturally occurring acaricides that may serve as an alternative to synthetic acaricides. Although some naturally occurring compounds are considered toxic to the environment, the soft acaricides oxalic acid, thymol, and formic acid 65% are usually safe for honey bee colonies and beekeepers, when handled appropriately. The current study investigated the effectiveness of formic acid (10, 15, and 20 mL/hive), oxalic acid (4.2, 3.2, and 2.1%/hive), and thymol (6, 4, and 2 g/hive) in controlling mite infestation. The results indicated that all treatments significantly reduced the mite population (p < 0.05). The average efficacies of oxalic acid at 3.2% (94.84% ± 0.34) and 4.2% (92.68% ± 0.37) were significantly higher than those of the other treatments. The lowest efficacy was recorded in formic acid 65% at 10 mL (54.13%). Overall, the results indicated that soft acaricides-such as oxalic acid at 3.2% and 4.2% concentrations-are very effective at controlling varroa mites and can be used in broodless conditions without side effects.

BACKGROUND: Chemotherapy for rhabdomyosarcoma (RD) is effective, but it has critical side effects and unavoidable challenges. Photodynamic therapy (PDT) is an approach to treating cancer with relatively moderate side effects. Plant products are a rich source of polyphenols, which have potent antioxidant and anticancer activities. Therefore, their research has become an emerging field in recent decades. PURPOSE: This work aimed to evaluate the potential of hydrophobic extract of Ficus Carica (FC) to determine whether FC in the presence of low dose chemo and Aluminium Phthalocyanine (Photosense®) mediated photodynamic therapy synergistically enhances the treatment efficacy of RD cells. METHOD: FC with and without combination with individual therapeutic modalities like photosense mediated photodynamic therapy, chemotherapy, and their combinations were studied for cell viability and morphological changes in invitro RD cells. A semiconductor diode laser (630 nm) was used as a light source in PDT. The cytotoxic effect of FC on cell viability and cellular morphological changes were investigated by MTT reagent and a camera attached to an inverted visible light microscope. The effect of FC, followed by di-combination with low dose chemo (doxorubicin-HCl, and dacarbazine), Photosense® mediated PDT and chemo-Photosense® mediated PDT (tri-combination) at 630 nm diode laser and 10 J/cm2 fluency were also investigated by MTT reagent. The combination index method is used to identify the synergistic effect of combination therapy by using CompuSyn software based on the Chou-Talalay method. RESULTS: The dose-dependent effect of FC on cell viability and cellular morphological changes were observed in the RD cell line. It was found that the pre incubation of FC potentiated the anticancer effect as a neoadjuvant agent for doxorubicin-HCl and decarbazine based chemotherapy, Photosense® mediated PDT and chemo-PDT (tri-combination) with synergistic effect (CI<1). CONCLUSION: These results suggest a possible thread that the low dose combination of the aforementioned therapeutic modalities in the presence of FC remarkably enhances the treatment efficacy of RD in comparison with a single-agent treatment modality. The proposed sequence of FC with chemo and PDT might present better therapeutic outcomes in RD therapies and may provide result for RD metastasis. FC may also be used in the application of phyto-PDT to cancer in the future.

Urea is the most popular and widely used nitrogenous fertilizer. High soil urease activity rapidly hydrolyses applied urea to ammonia which contributes to soil nitrogen (N) losses and reduces N use efficiency of crop plants. The ammonia losses can be minimized by the inhibition of soil urease activity which has been explored using various potential chemical inhibitors. However, the soil urease activity inhibition potential of plant extracts is rarely explored to date. In the present study, extracts of 35 plant materials were taken and evaluated against jack bean urease. Eleven extracts, showing >50% jack bean urease inhibition, were selected and further investigated in 13 soils collected from various districts of Punjab, Pakistan. Interestingly, except Capsicum annum, Melia azedarach, Citrus reticulata and Quercus infectoria, the plant extracts showed urease inhibition activities in soils, the extent of which was lower as compared to that observed in jack bean urease though. Maximum urea hydrolysis inhibition (70%) was noted with Vachellia nilotica which was 40% more than that of hydroquinone (50%) followed by that of Eucalyptus camaldulensis (24%). The extracts of V. nilotica and E. camaldulensis were coated on urea and applied to soil in the next step. At 21st day, 239% and 116% more urea-N was recovered from soil treated with V. nilotica and E. camaldulensis extracts coated urea, respectively, as compared to uncoated urea. Conclusively, these results indicated that the coating of V. nilotica and E. camaldulensis extracts on urea prills prolonged urea persistence in soil owing to minimum urea hydrolysis, probably, the extracts of V. nilotica and E. camaldulensis showed their urease inhibition potential. The results of this study provide a base line for the identification of new soil urease inhibitor compounds from plant materials in future.

Bergenia ciliate (B. ciliate) leaf extract was used as a reducing and stabilizing agent for the synthesis of silver-copper oxide nanocomposite (Ag-CuO NC). Scanning and transmission electron microscopies (SEM and TEM) were used to examine the structural morphology, and the average particle size was determined to be 47.65 nm. The phase confirmation and crystalline structure were examined through the X-ray diffraction (XRD) technique, where cubic and monoclinic geometries were assigned to Ag and CuO. The energy dispersive X-ray (EDX), Fourier transform infrared (FTIR) and ultra-violet and visible (UV-Visible) spectroscopies were operated to analyse the elemental composition, functional groups and light absorption phenomena of the Ag-CuO NC. Under the full light spectrum, the photodegradation of Rhodamine 6G was recorded, and 99.42 percent of the dye degraded in 80 min. The Agar well diffusion method was followed to perform antibacterial activity against selected pathogens, and the activity was found to increase with increasing concentration of Ag-CuO NC. The ABTS free radical scavenging activity suggests that the activity of Ag-CuO NC is higher than ascorbic acid.

Ajuga bracteosa Wall. ex Benth. is an endangered medicinal herb traditionally used against different ailments. The present study aimed to create new insight into the fundamental mechanisms of genetic transformation and the biological activities of this plant. We transformed the A. bracteosa plant with rol genes of Agrobacterium rhizogenes and raised the regenerants from the hairy roots. These transgenic regenerants were screened for in vitro antioxidant activities, a range of in vivo assays, elemental analysis, polyphenol content, and different phytochemicals found through HPLC. Among 18 polyphenolic standards, kaempferol was most abundant in all transgenic lines. Furthermore, transgenic line 3 (ABRL3) showed maximum phenolics and flavonoids content among all tested plant extracts. ABRL3 also demonstrated the highest total antioxidant capacity (8.16 ± 1 µg AAE/mg), total reducing power, (6.60 ± 1.17 µg AAE/mg), DPPH activity (IC50 = 59.5 ± 0.8 µg/mL), hydroxyl ion scavenging (IC50 = 122.5 ± 0.90 µg/mL), and iron-chelating power (IC50 = 154.8 ± 2 µg/mL). Moreover, transformed plant extracts produced significant analgesic, anti-inflammatory, anticoagulant, and antidepressant activities in BALB/c mice models. In conclusion, transgenic regenerants of A. bracteosa pose better antioxidant and pharmacological properties under the effect of rol genes as compared to wild-type plants.

Optical properties can provide rich information about morphology and structure of tissues. Fresh and frozen muscle tissue samples of goat are investigated using imaging polarimetry to understand its structural nature. The outcomes demonstrate that the muscle tissues lose, to some extent, their integrity and organization on freezing. The fresh tissues offer very small circular retardance as compared to frozen samples. However, linear retardance is the main contributor in fresh muscle samples. Ultimately, linear and circular retardance can be used to differentiate fresh and frozen tissues. These investigations illustrate the capabilities of optical polarimetry for the characterization of muscle tissue structures. Specifically, the structure of biological tissue samples can be differentiated using real-time, cost effective and non-invasive optical polarimetry in the field of meat industry and biomedical diagnosis.

BACKGROUND: Diabetes mellitus is a chronic disease characterized by hyperglycemia that may occur due to genetic, environmental or lifestyle factors. Natural remedies have been used to treat diabetes since long and many antidiabetic compounds of varied efficacies have been isolated from medicinal plants. Rhazya stricta has been used for decades for the treatment of diabetes mellitus and associated ailments. Considering the folkloric use of R. stricta against diabetes, it was aimed to investigate the effectiveness of its root extracts against diabetes through in vitro assays and in vivo studies using animal model along with phytochemical profiling through GCMS. METHODS: Various fractions of Rhazya stricta obtained through column chromatography were evaluated for a variety of assays including α-glucosidase, Dipeptidyl peptidase-IV (DPP-IV), ß-secretase and Glucagon-like peptide-1 (GLP-1) secretion studies. For the in vivo studies the alloxan-induced diabetic mice were treated with root extracts and blood glucose levels, HbA1C, and other biochemical markers along with the histological study of the liver were done. The phytochemical identification was performed using an Agilent 7890B GC coupled to a 7010 Triple Quadrupole (MS/MS) system. GraphPad Prism software version 5.01 was used for statistical analysis. RESULTS: Majority of the extract fractions showed excellent results against diabetes by inhibiting enzymes DPP-IV (Up to 61%) and ß-secretase (Up to 83%) with IC50s 979 µg/ml and 169 µg/ml respectively with increase in the GLP1 secretion. The results of in vivo studies indicated a marked reduction in blood glucose and HbA1c levels along with positive effects on other parameters like lipid profile, liver functions and renal functions of extract-treated mice as compared to control. The histological examination of the liver demonstrated hepatoprotective effects against diabetes led changes and various classes of phytochemicals were also identified through GCMS in different fractions. CONCLUSION: The results revealed strong antidiabetic activity of R. stricta root with the potential to protect body organs against diabetic changes. Moreover, a variety of phytochemicals has also been identified through GCMS that might be responsible for the antidiabetic potential of Rhazya stricta root.

The present study aimed to screen the Rhazya stricta Decne root for its antihyperglycemic and antioxidants potential through invitro assays along with phytochemical and elemental analyses. The crude extract was prepared through maceration and fractionated using solvent-solvent extraction technique. The spectroscopic studies indicated the presence of various phytochemical classes in the extract and its fractions. The antioxidant assays showed notable results along with a good concentration of phenolic and flavonoid contents. Enzyme inhibition assays demonstrated glucose-lowering effects by inhibiting the enzyme activity which could reduce post-prandial blood glucose level. The Dipeptidyl peptidase-IV (DPP-IV) inhibition assay results showed the novel DPP-IV inhibition activity of the plant extract and all fractions showed noteworthy enzyme inhibition and antihyperglycemic activity. Conclusively, the Rhazya stricta root extract displayed its antioxidant and antihyperglycemic potential due to the presence of various classes of phytochemicals and micro-nutrients.


El presente estudio tuvo como objetivo examinar la raíz de Rhazya stricta Decne por su potencial antihiperglicémico y antioxidante a través de ensayos in vitro junto con análisis fitoquímicos y elementales. El extracto crudo se preparó por maceración y se fraccionó usando una técnica de extracción solvente-solvente. Los estudios espectroscópicos indicaron la presencia de varias clases fitoquímicas en el extracto y sus fracciones. Los ensayos antioxidantes mostraron resultados notables junto con una importante concentración de contenido fenólico y flavonoide. Los ensayos de inhibición enzimática demostraron efectos reductores de la glucosa al inhibir la actividad enzimática que podría reducir el nivel de glucosa posprandial en sangre. Los resultados del ensayo de inhibición de Dipeptidyl peptidase-IV (DPP-IV) mostraron la nueva actividad de inhibición de DPP-IV del extracto de la planta y todas las fracciones mostraron una notable inhibición enzimática y actividad antihiperglicémica. En conclusión, el extracto de raíz de Rhazya stricta Decne mostró su potencial antioxidante y antihiperglicémico debido a la presencia de varias clases de fitoquímicos y micronutrientes.

Embryonic stem cells provide an ideal system to study various therapies for serious human diseases such as juvenile diabetes, neurodegenerative diseases, heart diseases and cancer. Synthetic or natural compounds that affect cell proliferation and/or differentiation of embryonic stem cells are of great value. Focus of the current project was upon the isolation and evaluation of natural components from a medicinal plant; Rhazya stricta on proliferation/ differentiation potential of embryonic stem cells. For this purpose, after a series of fractionation and purification steps, 7 compounds named as RS1-RS7 were isolated from aerial parts of the plant. The effects of these compounds were evaluated on the morphology and rate of cell proliferation of mouse naive embryonic stem cells. Only RS7 inhibited the proliferation of cell and reduced the induction of differentiation of cell. The qPCR analysis confirmed that the expression of the selected pluripotency markers (Oct4, Nanog and Sox2) was down regulated by RS7 treatment as compared to control. Furthermore, upon withdraw of Leukemia inhibitory factor (lif) from medium; effect of RS7 to promote differentiation was enhanced. Through structure elucidation studies, RS7 was found to be ursolic acid. This study first time shows the effect of natural compounds of Rhazya stricta Decne. on mouse embryonic stem cells.
